Hypocretin/Orexin, Sleep and Alzheimer’s Disease

Advances in translational research provide key opportunities to explore the physiological and pathological effects of sleep different neurodegenerative diseases. Recent findings suggest that sleep-wakefulness dysfunctions may predispose disorders such as Alzheimer’s disease (AD), vice versa. New theories on link between β-amyloid tau secretion, accumulation clearance, its interaction with hypocretins/orexins (key neuropeptides regulating wakefulness) mechanistic ways better understand impact alterations pathogenesis AD. Further studies should validate whether changes circadian rhythm patterns could be used for early AD diagnosis prognostic markers cognitive decline. Longitudinal are needed, not only these biomarker interactions determine cause-effect relationship role behavior regulation amyloid plaque neurofibrillary tangle formation, but also identify best therapies related preventive strategies
